Loop-mediated Isothermal Amplification Test for Rapid Identification of Clinical Nocardia Isolates

Background: Nocardia is a Gram-positive and partially acid-fast bacterium. The species are widely distributed in the environment cause severe human infections. Nocardiosis not easily identifiable due to lack of pathognomonic clinical signs. Objectives: present study was designed develop evaluate simple quick method based on loop-mediated isothermal amplification (LAMP) assay for detecting spp isolated from bronchoalveolar lavage (BAL) samples. Methods: In this cross-sectional study, 357 BAL samples were collected two teaching hospitals. polymerase chain reaction (PCR) performed using set species-specific primers 16S rRNA gene. Kinyoun staining culture done Sabouraud dextrose plate. optimal LAMP condition at 65°C 45 min, with recognition limit as 1 pg DNA/tube 100 CFU/reaction. addition calcein manganous ions, agarose gel electrophoresis used visualize amplified products. Results: Out samples, 0 (0.0%), 4 (1.1%), 9 (2.5%), 10 (2.8%) strains identified by direct partial acid-fast, streak plate, PCR, methods, respectively.
